報告人簡介:Jean-Louis Mergny教授,現(xiàn)任職于巴黎綜合理工學(xué)院,光學(xué)和生物科學(xué)實驗室主任。2019年入選歐洲科學(xué)院院士和法國生物化學(xué)和分子生物學(xué)學(xué)會會士,也是法國國家健康與醫(yī)學(xué)研究院院士。Mergny教授主要從事特殊DNA結(jié)構(gòu)的生物物理化學(xué)性質(zhì)及在納米技術(shù)中的應(yīng)用研究。曾獲法蘭西科學(xué)院“Labbey”獎(2000),法國生物化學(xué)與分子生物學(xué)會“Maurice Nicloux” 獎(2001),法國國家健康與醫(yī)學(xué)研究院“Therapeutical research”獎(2002)。在Science, Nature Medicine, Nature Chem. Biol. 等期刊發(fā)表論文200多篇,他引超過40000次,h-index 96。
內(nèi)容摘要:Beyond the iconic double-helix, DNA and RNA are prone to structural polymorphism. I have dedicated my whole career to these “weird” nucleic acid structures. For example, we are developping tools to study the folding and polymorphism of unusual nucleic acid structures and to understand how they can compete with regular duplex-formation. G-quadruplexes (G4s), which are often found in aptamer sequences, are involved in key biological processes, including transcription, and can contribute to tumorogenesis. We designed and applied an algorithm for the prediction of G4 propensity to a variety of genomes, including cancer genomes, and then used a variety of biochemical and in cell techniques to confirm these predictions in a variety of organisms.
